HEMOGLOBIN is a dried preparation of bovine erythrocytes. It forms a stable solution at 2% after sterilization.
It is used as an enrichment substance in certain culture media such us Trypticasein and phosphate broth, to isolate by hemoculture fastidious germs as Haemophilus, Streptococcus, etc... and specially to prepare the Chocolate Agar Media, widely used on the isolation of pathogenic Neisseria, Gonococcus and Meningococcus.
Generally, the basic media are prepared separately at double concentration, just like the hemoglobin suspension.
It is sterilized and mixed at equal volumes, being the concentration of the complete medium reducted to a normal level
Packaging
Suspend 2 grams of the dehydrated haemoglobin in 100 ml. of distilled water. Mix the haemoglobin with 2 or 3 ml. of water until a paste is obtained. Gradually add the rest of the water until a homogeneous suspension os obtained . Sterilize in autoclave at 121ºC for 15 minutes Cool to about 50ºC and add the same quantity of sterile medium (GC Agar Base Columbia) in double concentration.
NOTA: To prepare Chocolate Agar add 2 ml. Of the Polyenrichement to 100 ml. Of GC Agar Base in double concentration and 100 ml. Of hemoglobin solution.
